Purification and characterization of potato lectin.

چکیده
Potato lectin (Solanum tuberosum agglutinin, STA), purified by affinity chromatography on tri-N-acetylchitotriose-Sepharose 6B, has Mr approximately 100,000, as estimated by gel filtration on Sephadex G-150 and is an aggregating system with a monomer Mr = 54,000, as estimated by sedimentation equilibrium analysis. Equilibrium dialysis showed that STA (dimer) has two binding sites for a specific sugar per molecule. STA has a high content of sugar, most of which is L-arabinose, and is rich in Hyp and Cys. On interaction with specific sugars, STA induced a UV difference spectrum having positive peaks at 292 and 285 nm characteristic of tryptophyl residues. The association constants with chitin oligosaccharides, determined from the intensities of the difference spectra at various concentrations of sugars, increased with increasing chain length of the sugar. Association constants obtained by frontal affinity chromatography of chitin oligosaccharides with STA-Sepharose were in good agreement with those obtained by difference spectra, whereas the association constants obtained by frontal affinity chromatography of STA with di- and tri-N-acetylchitotriose-Sepharose were much higher, presumably owing to the effect of multivalency of ligands. The CD spectra of STA in the far UV region indicate the presence of 40% of beta- and 60% of unordered form, and no alpha-helix conformation, which supports the structure suggested by the amino acid composition and the high content of sugar.

Background: The ethno-medical significance of Clerodendrum genus raises the interest towards the characterization of its seed lectin by inexpensive and most effective technique. Objective: The focus of this study is the purification, characterization, and evaluation of the antioxidant and antiproliferative potential of a galactose-specific lectin from Cler...
متن کاملPurification of a novel chitin-binding lectin with antimicrobial and antibiofilm activities from a bangladeshi cultivar of potato (Solanum tuberosum).
A new chitin-binding lectin was purified from a Bangladeshi cultivar 'Deshi' of potato (Solanum tuberosum L.) through anion-exchange and affinity chromatographies using a chitin column. Sodium dodecyl sulphate-polyacrylamide gel electrophoresis (SDS-PAGE) showed the molecular mass of the lectin as 20,000 Daltons. This molecular mass was almost half of the molecular masses of chitin-binding lect...
متن کاملPurification and Characterization of a Mucin Specific Mycelial Lectin from Aspergillus gorakhpurensis: Application for Mitogenic and Antimicrobial Activity
BACKGROUND Lectins are carbohydrate binding proteins or glycoproteins that bind reversibly to specific carbohydrates present on the apposing cells, which are responsible for their ability to agglutinate red blood cells, lymphocytes, fibroblasts, etc. Interest in lectins has been intensified due to their carbohydrate specificity as they can be valuable reagents for the investigation of cell surf...
متن کاملPurification, characterization and antifungal activity of LPA a lectin from Pachira aquatica seeds: their possible role in plant defence
Background The lectins are an important class of proteins ubiquitous in nature, which plays different roles and functions in biological processes such as recognition molecules within the immune system in animals and as storage proteins or in defence mechanisms against pest and pathogens in plants. Oliveira et al. [1] showed that rats fed on a lectin-rich diet from Pachira aquatica seeds had a p...
متن کاملThe purification and properties of the lectin from potato tubers, a hydroxyproline-containing glycoprotein.
1. Potato lectin has been purified and shown to be a glycoprotein containing about 50% of carbohydrate. Most of the sugar residues (92%) are arabinose; small amounts of galactose, glucose and glucosamine are also present. 2. The most abundant amino acid is hydroxyproline (16% of the residues), 11.5% of the residues are half-cystine and phenylalanine is absent. The lectin also contains about one...
